چکیده

A6-amino acid peptide fragment of the azurin molecule was observed to act on HCT116 cancerous cells and cell death was brought by apoptosis in vitro. Integrity of the cell membrane is an indispensable prerequisite for preserving the biological characteristics of a cell. In this study, changes in the morphology and ultrastructure of HCT116 (human colon carcinoma) was studied by atomic force microscopy (AFM) and Scanning Transmissiom Electron Microscope (STEM) before and after treatment with the azurin derived hexapeptide. In response to hexapeptide addition, the microstructure of the cell membrane of HCT116 cell displayed changes. With increase in reaction time, the degree of morphological changes on the surface of cell membrane increased. These changes included increase in the fluctuation of the surface components of the cell membrane, and roughness of cell. Cell viability was done by MTT assay. Analysis of the cell membrane root-mean-square (RMS) roughness showed that under the action of hexa peptide, RMS values for the cell membranes of HCT 116 cell type increased with time thus positively correlating to the reaction time. This work is of high significance for the visual diagnosis of early stage of apoptosis in colon cancer cells in response to azurin derived hexapeptide, The use of AFM and STEM can be a rapid and sensitive visual method for studying the interaction between cells and drug.

An answer to colon cancer treatment by mesenchymal stem cell originated from adipose tissue

Objective(s): Colon cancer is risen up with its complex mechanism that directly impacts on its treatment as well as its common prevalence. Mesenchymal stem cells (MSCs) have been considered as a therapeutic candidate for conventional disease including cancer. In this research, we have focused on apoptotic effects of adipose tissue-derived MSCs in colon cancer.Materials and Methods: MSCs were ob...

Expression of Recombinant Coagulation Factor IX in Human Amniotic Membrane-derived Mesenchymal Stem Cells: A New Strategy to Gene Therapy of Hemophilia B

Background: Hemophilia B is an X-linked hereditary disorder of blood coagulation system which is caused by factor IX (FIX) deficiency. Factor IX is a plasma glycoprotein that participates in the coagulation process leading to the generation of fibrin. Replacement of factor IX with plasma-derived or recombinant factor IX is the conventional treatment for hemophilia B to raise the factor IX le...

Mesenchymal Stem Cells Trigger Epithelial to Mesenchymal Transition in the HT-29 Colorectal Cancer Cell Line

Background and Objective: Mesenchymal stem cells (MSCs) promote metastasis in colorectal cancer; however, the mechanism underlying this process is not fully understood. Epithelial to mesenchymal transition (EMT) is a key step in tumor acquisition of metastatic phenotype. We aimed to investigate the effect of MSCs on the expression of EMT markers, as well as cancer stem cell markers in HT-29 col...
